| The European Pharmacopoeia (EP) is an international regulation used in the European region. It is recommended to use Norleucine (Nle) as an internal standard for the amino acid analysis listed in the EP. Moreover, the Resolution of Isoleucine (Ile) and Leucine (Leu) is required 1.5 or higher for system suitability.As an EP-compliant analytical method, we have developed a PH-Nle high-resolution method in which Nle can be used as an internal standard and the Resolution of Ile and Leu is 1.5 or higher.The PH-Nle High-resolution analysis method is suitable for precise quantification, which enables amino acid analysis with high-resolution even for components other than Ile / Leu. In addition to EP, this analysis method also complies with COMMISSION REGULATION (EC) No 152/2009 and ISO 13903-2005 Animal feeding stuffs, which stipulate analysis methods for feed (including pet food). | |

| In this report, we will introduce the PH-Nle High-speed analysis method that shortens the analysis time of the PH-Nle High-resolution analysis method (refer to AS/AAA-029).The PH-Nle High-speed analysis method can separate Norleucine (Nle), which is used as an internal standard substance, and analyze the Resolution of Isoleucine (Ile) and Leucine (Leu) is 1.5 or higher in 45 minutes.Since there are some peeks which the degree of resolution is inferior to that of the PH-Nle High-resolution analysis method, it may not be suitable for precise quantification of samples. However, the analysis method, which performs standard quantification at highspeed, is very useful when the emphasis is on shortening the analysis time. | |

Staple grains refer to the staple crops rice, wheat, and corn, while miscellaneous grains are a general term for the small caryopsis-bearing crops of the Gramineae family, such as barnyard millet, foxtail millet, and millet. This report introduces examples of protein-constituting amino acid analysis of staple grain (Non-glutinous rice) and three types of miscellaneous grains (Foxtail millet, Pilled millet, and Japanese barnyard millet). The analysis was performed using LA8080 HIGH SPEED AMINO ACID ANALYZER (AminoSAAYA) with Protein Hydrolyzate Analysis Method. The results showed that miscellaneous grains are highly nutritious foods because they contains more protein-constituting amino acids than non-glutinous rice. Knowing the amino acid content of nutritional components through amino acid analysis is useful for the nutritional evaluation of foods. |

| Oxytocin is a peptide composed of 9 amino acids. One of the tests performed to evaluate the quality of peptide drugs such as oxytocin is the confirmation of the amino acid composition. Presented here is the analysis of oxytocin using the high resolution column for the Hitachi L-8900 high-speed amino acid analyzer. The amino acid composition of oxytocin was determined by using an analysis method that conforms to the Japanese Pharmacopoeia. The standard reagent oxytocin acetate was used as a model sample of the drug product. | |

The PH-Nle High-resolution analysis method (refer to AS / AAA-029E) was used to analyze the 18 components of protein hydrolyzed amino acids and norleucine (Nle) added as an internal standard at once. Norleucine (Nle) was added as an internal standard. Nle can be analyzed in any sample, and each aminoacid component can be analyzed with high resolution. The PH-Nle High-resolution analysis method is useful for amino acid analysis in pet food. | |

This report introduces an example of measuring free amino acids, which are involved in the taste and flavor, of staple grain (Non-glutinous rice) and three types of miscellaneous grains (Foxtail millet, Pilled millet, and Japanese barnyard millet) using LA8080 HIGH SPEED AMINO ACID ANALYZER (AminoSAAYA) . The measurement results showed that miscellaneous grains had a higher amount of free amino acids than Non-glutinous rice, and that they all contained high amounts of Asparagine (AspNH2), GABA (γ-aminobutyric acid, g-ABA), and Arginine (Arg). Measuring free amino acids is useful for evaluating the characteristics of food. |
